乳酸菌是公认安全的食品级微生物,胞外多糖是其重要的次生代谢产物,具有很多保健功能.本实验室前期对一株高产胞外多糖的马奶酒样乳杆菌ZW3进行了全基因组测序,得到一段14.4 kb的EPS基因簇.本实验从该基因簇中选取一个在菌体生长过程中表达量变化明显的基因WANG_1297作为研究对象,生物信息学分析表明,WANG_1297编码蛋白属于Fer4_NifH蛋白超家族,具有ATP结合结构域,同时还有eps_fam多功能位点,与荚膜多糖的生物合成相关,跨膜结构预测表明,其编码蛋白为胞内蛋白;实时荧光定量PCR实验表明,其在多糖合成旺盛阶段表达量最高;在大肠杆菌中克隆表达WANG_1297基因,通过酶联免疫实验,得出该蛋白不具有酪氨酸蛋白激酶活性,推测其是通过结合多糖复合物而实现多糖链聚合.
