Conserved dopamine neurotrophic factor (CDNF) is atrophic factor for dopamine neurons.Intrastriatal injection of CDNF was able to restore the dopaminergic function and prevent the degeneration of dopaminergic neurons in substantia nigra administered four weeks after 6-OHDA.In the present study, we delivered CDNF gene by recombinant adeno-associated virus AAV2.CDNF into the striatum of 6-OHDA lesion rats, and achieved a robust restoration of motor behavior.The recovery of the behavior lasted for at least 8 month.Tyrosine hydroxylase (TH)-positive neurons in the substantia nigra pars compacta (SNpc) and TH-positive fibers in the striatum were almost completely rescued.The reduction of DAT function in the striatum of 6-OHDA lesion rats was attenuated significantly as measured by PET.Our findings demonstrated the long-term neurorestoration after striatal infusion of AAV2.CDNF, and implied AAV2.CDNF might have potential as neurorestorative therapy of Parkinsons Disease.
