Autophagy is important in protein degradation.We have found that lysosomal cathepsins are important in degrading alpha-synuclein and mutant huntingtin which are the key proteins accumulated in Parkinsons and Huntingtons diseases and play key roles in disease pathogenesis.In mouse models, primary neuron cultures and cell line based models, cathepsin D activities are important for cell survival in response to increased alpha-synuclein and mutant huntingtin.These studies provide new insights into regulation of autophagy and protein degradation pathways and new potential for designing novel therapeutic approaches against neurodegeneration.
