Hippocampus-mediated rewiring of cortical microcircuitry underlies memory storage

  Sensory inputs engage selective responses in cortical neurons to generate the representation of the external world.However,the way that such selectivity is formed and modified by experience is largely unknown.In vivo imaging of cortical cellular activity revealed the multilaminar structure of memory processing.Network perturbation by targeted cell ablation indicated that the excitatory components in the microcircuit generate selective responses in their downstream,while the inhibitory components govern the downstream activity levels.Furthermore,the learningconditioned cortical microcircuit across layer 4 to layer 2 underlies the neuronal activity dynamics,showing a local signal processing rule.Importantly,learning changed the observed activity correlations,implicating memory-related rewiring.Lesion of hippocampus after learning prevented the formation of new context-specific circuits.By revealing the structure and rewiring of the learning-conditioned microcircuits,our work offers novel insights into the cortical storage of specific long-term memory.
