Siderophores are important organic substances for iron chelation in microorganisms.Intracellular siderophores have been proposed to play a role in iron storage and recognized as asexual spore germination factors for several microorganisms.Both of sidC and sidD were reported to be related with nonribosomal peptide synthetases (NRPS) involved in biosynthesis of ferricrocin (FC) and triacetylfusarinine C (TAFC).Genomic DNA of Chinese native grass endophytes including 26 Epichlo(e) spp strains and 30 Neotyphodium spp.strains were extracted by IQS (Improved quick and safe method) method.Degenerate PCR primers were designed based on the conservative amino acid sequences of sidC and sidD through CODEHOP website.Approximately 750bp and 1200bp fragments were obtained through degenerate PCR when DNA from strains Rnj0119 and Rnj0102, which were used as the template, and sequenced respectively.After blasted in NCBI, these two DNA fragments were recognized to be highly similar with sequences of sidC and sidD fragments respectively.These two fragments distributed widely in all other tested 55 strains.Phylogenetic analyses basing on sidD fragments from strains used in this study exhibited that these strains clustered with foreign strains with bootstrap value of 86%.SidD fragment of 12/15 strains contained about 171~951 bp of an open reading frame (ORF), and an adenylation domain (A domain) of about 344~780 bp.Through a chrome azurol S (CAS) assay, media color of tested strains tumed from blue to pink or orange, indicating the productivity of triacetylfusarinine C by these Chinese native grass endophytes.This also demonstrated that CODEHOP could be simple and feasible to design degenerate primers for detection of genes from unknown strains, and this primers designing method can be used to explore the presence of the other NRPS genes in fungal genomes.The test strains have the potential ability to synthesis different hydroxamate peptide siderophores.SidC and sidD could be two new NRPS genes in endophytes.
