Objective: To evaluate the effect of epigallocatechin-3-gallate (EGCG) on microcystin LR (MC-LR) induced liver injury of mice.Methods: A total of 50 male mice were randomly divided into five groups.Mice in control group and MC-LR group were given 0.9% saline solution intragastric administration(i.g), in EGCG groups were given 10 mg/(kg.d)、 25 mg/(kg.d)、 50 mg/(kg.d) EGCG i.g prior to MC-LR intoxication consecutively for 21 days.Mice in MC-LR group and EGCG groups received continual intraperitoneal injections of MC-LR at dose of 10 μg/(kg.d) for 11 days from day 11 till sacrifice.The liver histopathology, alanine aminotransferase(ALT), aspartate aminotransferase(AST), liver antioxidant enzyme and lipid peroxidation product were evaluated immediately.Results: Given EGCG treatment can reduce MC-LR induced liver pathological injury.Compared with control, the ALT, AST and MDA level were significantly increased (P<0.05), the SOD level were significantly decreased (P<0.05) in MC-LR group and EGCG groups.High and medium dose of EGCG pretreatment significantly inhibited MC-LR-induced ALT and AST raised(P < 0.05).The significant differences in SOD and MDA level were found in all EGCG pretreatment groups with a dose-effect relationship when compared with MC-LR group.(P < 0.05).Conclusion EGCG can reduce the damage in liver histopathology and liver function induced by MC-LR.It also can reduce the level of lipid peroxidation, increase antioxidant enzyme activity of nepatocyte,then show a certain effect on mitigating the liver injury induced by microcystin LR of mice.
