Recent introduction of lmatinib Mesylate (IM) and other tyrosine kinase inhibitor (TKI) therapies has had a major impact on treatment of chronic myeloid leukemia (CML) worldwide. However, TKI monotherapies are not curative and initial and acquired TKI resistance, as well as relapse, remain significant challenges. We have recently demonstrated that CML stem cells are less responsive to IM and other TKIs and are a critical target population for TKI resistance. Therefore, improved treatment approaches to prevent the development of resistant subclones, by targeting other key molecular elements active in CML stem cells, are thus clearly needed.
