Objective: To investigate the effect of fibroblast growth factor-21(FGF-21)on triglyceride(TG)of nonalcoholic fatty liver disease(NAFLD)in vivo and vitro.Methods:(1)Free fatty acids(FFA)mixture was used for model building of steatosis in vitro.Then LO2 cells were treated with FGF-21 or fenofibrate.After 24 hours,Oil red O staining was performed to observe the degree of steatosis and intracellular TG contents were detected.The mRNA and protein of SREBP-1c were examined by RT-PCR and Western blot.(2)High fat diet(HFD)was used to establish the animal model of NAFLD.HFD mice were treated with FGF-21 or fenofibrate for 8 weeks.The liver samples were stained with HE.Meanwhile,levels of serum TG,ALT,AST and TG contents of liver samples were measured.
