  Jasmonic acid(JA)plays an important role in various aspects of plant development and responses to stresses,however,it is so far unknown how JA biosynthesis is transcriptionally modulated in relation to JAmediated cold response in economically important fruits.Here,we report the possible involvement of a Group I WRKY transcription factor(TF)MaWRKY26 in cold stress response of banana fruit.MaWRKY26has two WRKY domain that contains the highly conserved amino acid sequence WRKYGQK and two putative zinc finger motif.Subcellular localization and transient expression analysis showed that MaWRKY26 was localized to the nucleus and possessed transcriptional activation activity.Quantitative realtime PCR(qRT-PCR)indicated that MaWRKY26 was cold-and MeJA-inducible.Moreover,yeast-one hybrid(Y1H)and electrophoretic mobility shift(EMSA)assays demonstrated a specific binding of MaWRKY26 to promoters of three JA biosynthesis genes MaLOX2,MaAOS3 and MaOPR3.Interestingly,further yeast two-hybrid(Y2H),bimolecular fluorescence complementation(BiFC)and co-immunoprecipitation(ColP)investigations showed that MaWRKY26 interacted in the nucleus with a VQ motif-containing protein MaVQ5,and their interaction attenuated the transactivation of WRKY26 to JA biosynthesis genes.These results provide clear evidence that the MaVQ5 protein acts antagonistically with cold-responsive MaWRKY26 in the regulation of JA biosynthesis genes.Taken together,our findings reveal new information in helping to understand the transcriptional regulatory of JA biosynthesis in response to cold stress.
