Dietary carotenoids are essential and important for humans because mammals are incapable of de novo synthesis of vitamin A,but wheat cultivars,as the main crop, have low carotenoid contents. In order to demonstrate and modify the complex carotenoid metabolic pathway in wheat grains and to improve the nutritional value of wheat,we used combinatorial genetic transformation approach to transfer the bacterial phytoene synthase gene (CrtB),carotene desaturase gene (Crt1),lycopene beta cyclase (CrtY) and Beta carotene hydroxylase (CrtZ) controlled by endosperm-specific 1DX5 promoter into the common wheat cultivar Bobwhite. We obtained 1280 regenerated seedlings by use of particle bombardment method.Identification and characterization of positive transgenic lines are underway. This strategy may allow us to generate transgenic plants with extraordinary high levels of β-carotene and other carotenoids.
