Mesoporous silicon nanomaterials due to the huge specific surface are, pore size adjustable, easy to modify, higherbiocompatibility characteristic, more and more received the attention of people.Especially, as a genetic drug carrier has the incomparable superiority than other carriers,with the development of nanoscience, nanomaterial will hew out a new road in the disease treatment.This experiment, first synthetic MSN mesoporous silicon nanoparticles, which through calcined, amination, carboxyl modify, after modified nanoparticles pass a series of physical and chemical characterization,such as SEM, TEM, Zeta potential, XRD, BET, BJH.And then different modified nanoparticles used for siRNA adsorption, release and protection performance, and based on trace spectrophotometer and polyacrylamide gel electrophoresis to analysis the result.Finally siRNA adsorption of different nanoparticles transfection GFP-MCF7 cells, through the confocal fluorescence microscope and flow cytometry observation intake condition and corresponding gene silence efficiency (RNAi).In addition,using the MTT method,determination ofnanoparticles for the cell toxicity, through the above experiments for mesoporous silicon nanometer material in clinical applications provide beneficial basis.
