A method was established for determining Rutin [1],Troxerutin,Baicalin and Quercetin [2] by HPLC.The analysis was performed on Shim-pack VP-ODS(150 mm × 4.6 mm,5 μm) column with mobile phase contained phosphoric acid (0.5%)-methanol (63 :37).The flow rate was 0.8 mL.rain"1,the detection wavelength was set at 333 nm,the temperature of column was 40 ℃ and the injected volume was 20 μL.Under this condition,Rutin,Troxerutin,Baicalin and Quercetin could be completely separated in 35 minites.The lined range of four flavonoids [3] was between 1~10 μg and linear correlation coefficient (R2) was up to 0.9866.The relative standard deviations of Rutin,Troxerutin,Baicalin and Quercetin were 1.6%,2.1%,3.0% and 1.1% (n=6),respectively.The method was applied to determination of four flavoniods in Astragalus,the average recovery of four flavoniods was between 92.2%~109%.
