Tachyplesin I (TP-I) are a sort of cationic antimicrobial peptide (CAMP) which are composed of 17 amino acids residues extracting from horseshoe crabs blood cells.Previous study showed that TP-I possess broad-spectrum activities against Gram-positive and Gram-negative bacteria,fungi, viruses, and some cancer cell.High costs would limit the application of TP-I if use horseshoe crabs blood and synthetic as source of TP-1.Hence, Genetic engineering is the most promising way to mass produce TP-I.Although the research of genetic engineering expression of TP-I began in the end of the 90s, but there has been no substantive progress.One of the most important reason is that expression host cannot overcome the kill effect of TP-I, leading to express quantity could not reach the requirement of industrialization.To crack this problem, we have bred a TP-I resistant Bacillus subtitles strain through long-term stress, and the resistant mechanism of B.subtitles has been preliminary explored.The purpose of this study is to study the resistant action of TP-I resistant B.subtitles strain to antibiotic and explore the correlation of B.subtitles resistance to TP-I and antibiotic, in order to further speculate the resistant mechanism of B.subtitles resistance.The resistance of long-term stressed B.subtitles to ampicillin, polymyxin B, bacitracin, vancomycin and actinomycin D were detected by K-B paper disk diffusion.The cell surface negative charge of B.subtitles is assessed by Cytochrome C method.Results showed that the resistance of stressed strains to ampicillin and bacitracin improved 128 times compared with wild strains, improved 32 times to vancomycin and actinomycin D, and improved 8 times to polymyxin B.Detection results showed that cell surface negative charge of B.subtitles reduce more than three times.In conclusion, B.subtitles evolved high resistance to other CAMPs or cationic antibiotics after long-stress by TP-I since its surface negative charge significantly reduced which can hinder CAMPs and cationic antibiotics into cell.
