Nuclear hormone receptors (NHRs) such as peroxisome proliferator-activated receptors (PPARα,PPARβ/δ,and PPARγ),liver X receptors (LXRα and LXRβ),and estrogen-related receptors (ERRoα and ERRγ)are involved in regulating lipid,cholesterol,carbohydrate and steroid metabolism by governing the expression of target genes as sequence specific transcription factors.Through their divergent ligand binding domains (LBDs),these receptors discriminate among specific ligands and differentially recruit coactivators or corepressors to regulate their transcriptional activities.Many natural products including those derived from Traditional Chinese Medicine (TCM) have been demonstrated to be beneficial for preventing coronary heart diseases and diabetes due to the abilities to lower plasma cholesterol,fat and glucose levels.In this presentation,I will present evidences that suggest certain natural products bind directly to NHRs to affect the expression of their target genes while others affect the activities of cofactors to modulate their activities.
