Atomic force microscopy(AFM) has supplied a powerful approach for the bioimaging applications since it can work in physiological environment to keep the natural structures of bio-samples.In the commercial AFM operational modes, including contact and tapping modes, the contact or intermittent contact between the tip and sample not only would damage the sharpness of the tip, but also deform or even damage the structure of the soft flexible bio-samples, which limits the applications of high resolution imaging on biological surfaces.Non-contact frequency modulation AFM (NC-FM-AFM) is one of the techniques for overcoming the limitation. The non-contact operation mode would prevent the sharp tip apex of only a few atoms from disintegrating during imaging and the surface of soft bio-samples from damaging. We have developed a novel NC-FM-AFM with low noise. In our systems, we utilized the capacitive force to excite the cantilever to eliminate the unwanted peaks in the spectrum induced by the coupling between the mechanical part of the liquid cell and the surrounding water, which increased the quality factor Q hence enhanced the force sensitivity.
