Alteration of integrinβ1 expression in spinal dorsal cord of CCI rat

　　Objective Neuropathic pain is associated with reorganization of spinal synaptic circuits, implying that adhesion proteins must be involved.The present study was to examine the alteration of adhesion protein integrinβ1 expression in relationship of neuropathic pain.Methods The left common sciatic nerve of SD rats was ligated to produce a chronic constrictive injury (CCI) model.Mechanical allodynia was assessed.The co-expression of integrinβ1 with neuron/glia cell was identified by double-labeling immunofluorescence, and the alteration of integrinβ1 expression was detected by RT-PCR, immunohistochemistry and immunoblot analysis.Results In L5, integrinβ1 is co-expression with neuron and glia cell within lamina Ⅱ.The mRNA and protein level of integrinβ1 undergoes a time-dependent increase across spinal dorsal cord after CCI.Conclusion These results suggest that integrinβ1 plays a crucial role in a painful peripheral neuropathy, making it a very promising target for the development of a novel class of analgesics.
