Proteins serve crucial functions in essentially all biological processes,but lack appropriate physical chemical properties and metabolic stability to be ideally suited as therapeutics.Here,I use an illustrative example to discuss the challenge inherent in the discovery of small-molecule mimics of sup.eroxide dismutase (SOD) as therapeutics.Many human diseases are associated with the overproduction of oxygen free radicals that inflict cell damages.Under normal circumstance,this radical burden is controlled by the SOD enzymes.Although protective roles of SOD enzymes have been shown in many diseases,such as myocardial ischemia-reperfusion injury,inflammation,etc,clinical trials of this enzyme had to be curtailed because of lack of oral availability,lack of access to intracellular space,immunogenic response,short half-lives in serum,etc.
