目的建立封闭群实验用小型猪遗传质量标准和遗传质量检测方法。方法根据群体遗传学理论,参照实验动物哺乳类实验动物的遗传质量控制(GB 14923-2001),在查阅大量文献资料的基础上,建立封闭群实验用小型猪遗传标准。检测方法采用微卫星标记分析技术。从GeneBank中挑选出的100个猪微卫星位点进行PCR扩增和条件优化,通过琼脂糖电泳、聚丙烯酰胺电泳和STR扫描三级筛选,根据位点在染色体分布情况和等位基因数目、多态性等优化出适于封闭群实验用小型猪遗传检测的微卫星位点组合,建立检测方法。结果初步建立了封闭群实验用小型猪遗传质量标准和检测方法。
OBJECTIVE To establish the genetic quality standard and genetic quality testing method for miniature pigs in closed herd experiment. Methods Based on the population genetics theory, referring to the genetic quality control of laboratory animals and mammals (GB 14923-2001), on the basis of reviewing a large amount of documents and materials, the genetic criteria of miniature pigs in experimental population were established. Detection method using microsatellite marker analysis technology. 100 pig microsatellite loci selected from GeneBank were used for PCR amplification and optimization of conditions. The results were analyzed by agarose gel electrophoresis, polyacrylamide gel electrophoresis and STR scanning. According to the distribution of chromosomes and the number of alleles, Polymorphism and other optimization of microsatellite loci combinations suitable for genetic testing of miniature pigs in a closed group experiment, the establishment of detection methods. Results The genetic quality standard and detection method of miniature pigs in closed-group experiment were initially established.
