目的:探讨地骨皮醇提液对H2O2诱导的HUVEC细胞凋亡的影响,探讨地骨皮醇提液对血管内皮的保护作用,为研究糖尿病并发症治疗的新药提供新的理论基础。方法:将HUVEC细胞分为正常对照组、高药物浓度组、H2O2组和药物预处理+H2O2作用组,作用18~24h后,用显微镜观察细胞形态改变,用流式细胞仪通过Annexin V/PI双染法检测各组细胞的凋亡百分比,用RT-PCR检测Bcl-2基因的表达变化。结果:地骨皮醇提液有效降低H2O2引起的细胞凋亡率;地骨皮醇提液可以增加凋亡相关基因Bcl-2的表达。结论:地骨皮醇提液对H2O2诱导的HUVEC细胞凋亡有不同程度的抑制作用,对血管内皮具有保护作用。
OBJECTIVE: To investigate the effect of Diterpenoid alcohol extracts on the apoptosis of HUVEC cells induced by H2O2, and to explore the protective effect of Dipidermal Derivatives on the vascular endothelium, which will provide a new theoretical basis for the study of new drugs for the treatment of diabetic complications. METHODS: HUVEC cells were divided into normal control group, high drug concentration group, H2O2 group, and drug pretreatment + H2O2 treatment group. After 18-24 hours, the morphology of cells was observed with a microscope and flow cytometry was used to pass Annexin V/PI. The percentage of apoptosis in each group of cells was detected by double staining, and the expression of Bcl-2 gene was detected by RT-PCR. RESULTS: Dioscorea bulb extract could effectively reduce the rate of apoptosis induced by H2O2. The extract of Dipisperm ethanol could increase the expression of apoptosis-related gene Bcl-2. Conclusion: Decoction of terbium epidermis can inhibit the apoptosis of HUVEC cells induced by H2O2 to varying degrees, and has protective effect on vascular endothelium.
