  Fluoride and sodium sulfite are two well known environmental contaminants, which pose a serious threat to male reproduction.The present study aims to determine the effects of exposure to fluoride and sodium sulfite on the mouse testis proteome as a first step toward the investigation of the potential damage mechanism in terms of MAPK signaling pathway.72 sexually male Kunming mice were randomly assigned to four groups (n =18) : Control group (given distilled water), NaF group (150mg NaF/L), Na2SO3 group (500mg Na2SO3/L) and Combination group (150mg NaF/L and 500mg Na2SO3/L).After 98 consecutive days, a two-dimensional gel electrophoresis approach was employed to investigate the changes in protein expression in the testes.Proteomic analysis was conducted using PDQuest 8.0 software and MALDI-TOF-TOF, which led to the identification of 19 differentially abundant proteins including 2 down-regulated proteins: A.14-3-3, plays a role in MAPK signaling pathway by recognition of a phosphoserine or phosphothreonine motif.B.peroxiredoxin-6, involved in redox regulation of the cell, may play a role in the regulation of phospholipid turnover as well as in protection against oxidative injury.And 17 up-regulated proteins included in inositol monophosphatase 1, ubiquitin C-terminal hydrolase L3, Chain n, sperm surface protein Spl7, cytochrome b-5, uncharacterized protein LOC691496, Prdx2 protein, Rho GDP dissociation inhibitor (GDI) alpha, ubiquitin carboxyl-terminal hydrolase isozyme L1, alpha-globin, Chain M, adenylate kinase isoenzyme 1 isoform 2, L-lactate dehydrogenase B chain, 14 ku phosphohistidine phosphatase, cytochrome c oxidase subunit 5A, and cytochrome b-cl complex subunit 1.Among these proteins, 14-3-3 exerted critical roles in the regulation of cellular phenomena in MAPK signaling pathway through phosphor-dependent binding to a lot of intracellular proteins that were targeted by various classes of protein kinases.Hence, the change of 14-3-3 expression possibly has an influence on the male reproduction, which must be vigorously further investigated.
