EFFECT OF DENV-2 INFECTION ON THE EXPRESSION OF IL-29 IN PRIMARY HUVEC WITH DIFFERENT PHYSIOLOGICAL

  Objective: Cultured of primary vascular endothelial cells from human umbilical veins endothelial cells (HUVECs) and simulation of substrate stiffness about HUVECs.To observe inoculated of primary HUVECs on hydrogel by dengue virus-Ⅱ(DENV-2) infected primary HUVECs which induced of IL-29.Methods: As to isolate and culture HUVECs, take experiment by us ing the well-cultivated fifth generation of HUVECs.Our laboratory had successfully established with hydrogel of 4Kpa and cells were inoculated on hydrogel, then infected the primary HUVECs by using the DENV-Ⅱ (MOI=10), then we detected the death rate and infection rate of the cells after 48 hours by using flow cytometry method, along with that, we also inspected the mRNA gene chip, at last, we tested the gene expression result through real-time fluorescent quantitative PCR and double antibody sandwich ELISA detected the difference of gene expression.
