目的:比较海藻玉壶汤(生甘草)组、海藻玉壶汤(炙甘草)组、去海藻(生甘草)组、去海藻(炙甘草)组、去甘草组、去海藻甘草组的小鼠急性毒性,评价海藻-甘草反药组合在复方中应用的毒性。方法:灌胃给与不同配伍组合不同剂量的海藻玉壶汤,计算出LD50,不符合LD50评价条件的组别,采用连续给药28 d,即累计半数致死量的方法来进行毒性评价;眼球采血,通过检测小鼠血清肝、肾功能相关指标评价其对小鼠肝、肾系统的影响。结果:各给药组均未得出LD50,海藻玉壶汤(生甘草)组LD50(16)为71.08 g·kg-1,药物剂量21 g·kg-1;海藻玉壶汤(炙甘草)组LD50(3)为68.28 g·kg-1,剂量21 g·kg-1;去甘草组LD50(12)为55.78 g·kg-1,剂量20 g·kg-1;去海藻(生甘草)组LD50(22)为72.27 g·kg-1,剂量21 g·kg-1;去海藻甘草组LD50(21)为79.62 g·kg-1,剂量18.1 g·kg-1;去海藻(炙甘草)组小鼠连续给药28 d仍未达到半数致死量,最大耐受量为生药80 g·kg-1,药物剂量20 g·kg-1;给药各组对小鼠的肝、肾脏功能均未见明显影响。结论:给药各组LD50(n)均属无毒的范畴,且对小鼠的肝、肾脏功能指标未见明显影响。
