The classical experiments of Anfmsens group demonstrating the oxidative refolding of denatured reduced pancreatic ribonuclease into active enzyme with correctly paired disulphide bonds established that functional higher-order protein structure can be ftmdamentally determined by primary structure (Anfinsen 1973).The history and recent progress of the field of the computational prediction of folding pathways and three-dimensional structure of proteins from their amino acid sequences has been elegantly summarised by ProfeSsor Harold Scheraga (Scheraga 2007).
