Apical extracellular matrix (aECM) plays a central role in epithelial tube geometry regulation.In the Drosophila tracheal system,a secreted chitin deacetylase Serpentine (Serp) expressed by the tracheal cells plays a key role in tube length restriction.Here we show that fly fat body,an equivalent mammalian liver,also transcripts serp mRNA and produces Serp protein.Fat body-produced Serp diffuse in haemolymph,taken up by the tracheal cells,and transcytosed to the lumen and functionally support normal tracheal development.This process becomes defective in mutants of shibire/dynamin,rab9,and shrub/vps32,leading to massive accumulation of Serp in the fat body.Our results reveal that the mesodermal organ fat body actively contributes to the tracheal development.Regulation of airway morphogenesis by the fat body may have an implication in matching the size of the circulatory system to the body dimension.
