The Androgen Signaling in Prostate Tumorigenesis and Prostate Cancer Progression

　　The androgen receptor (AR) mediates the biologic functions of androgens in prostate development, growth, function, tumorigenesis, and cancer progression.The general objective of our laboratory is to understand the detailed molecular mechanisms by which AR activates or represses key target genes that play critical regulatory roles in survival, differentiation, and proliferation of cells of the prostate, as well as in early events leading to prostate tumorigenesis.Toward this objective, we have identified a 44-kDa protein (p44) as a novel AR cofactor to regulate AR-driven gene expression.We further demonstrated that p44 in the cytoplasm was correlated with proliferation of prostate epithelial cells during prostate development and with initiation of prostate cancer.In contrast, the p44 in the nucleus inhibited epithelial cell growth and promoted cell differentiation.These observations suggest that p44 translocation from the nucleus into the cytoplasm is an essential step in initiating epithelial cell proliferation in the prostate gland.
