ADAM2是一种新型的癌/睾丸抗原(Cancer-Testis Antigen,CT抗原).本研究旨在利用基因重组方法制备ADMA2抗体,检测前列腺癌细胞系ADMA2的表达情况.PCR扩增编码人ADAM2第384-473位氨基酸的基因序列,构建PGEX-4T1/ADAM2原核表达质粒,诱导表达GST-ADAM2融合蛋白,制备多克隆抗体.利用该抗体检测ADAM2蛋白在人及小鼠前腺癌细胞系中的表达.发现低恶性度前列腺癌LNCaP、PC3细胞系中,ADAM2表达水平极低,而高恶性度PC3M、DU145细胞系中ADAM2表达量很高,提示了其在前列腺癌进展中潜在的重要功能.同时,小鼠前列腺癌RM1细胞系中ADAM2蛋白表达量高,与人前列腺癌细胞系相比,并能够检测到大小约30KD的剪切型,且与睾丸组织ADAM2的剪切型显著不同.结论：成功制备了ADAM2抗体,为进一步研究ADAM2蛋白在前列腺癌进展过程中的功能奠定基础.
