Humanin (HN) is a 24-aa peptide encoded by a newly identified gene cloned from an apparently normal brain region from patients with Alzheimers disease (AD).Studies on both cultured neurons and AD models in rats showed that HN could attenuate AD-related insults effectively.It was,therefore,considered as a selective neuroprotective peptide against AD.The mechanism underlying the neuroprotection of HN at molecular and cellular level,however,strongly suggested that HN play broad protective roles for neurons rather than just for AD-related insults.Firstly,neuronal loss could be neutralized by HN via elevating cellular ATP level.Next,neuronal apoptosis mediated by both intrinsic and extrinsic signal pathways could be attenuated by HN.Last,the Ca2+ overloading triggered by amyloid beta peptide could be inhibited by HN.We,therefore,hypothesized that HN is a neuroprotective factor against broad insults.The current research had investigated the neuroprotection of HN against NMDA-induced neurotoxicity,an in vitro paradigm of neurodegeneration representative of pathologically neurotoxic insults of excitotoxicity.
