基因治疗是向病变细胞中导入正常基因以替换和修正病变基因，从而达到治疗的目的。由于磷酸基团的存在，核酸表面是带电负性的，同时，细胞表面有很多带负电荷的蛋白以及糖脂化合物，从而导致核酸很难直接通过胞吞作用进入细胞，因此，核酸能否顺利的进入细胞成为基因转染的难点。
