Methyl-coenzyme M reductase(MCR)is a nickel-containing enzyme responsible for the last step of methanogenesis in methanogenic archaeabacteria.It is capable of catalyzing methane formation from methyl-coenzyme M(CH3-SCoM)and coenzyme B(CoB-SH),and also its reverse reaction(anaerobic methane oxidation).By our calculations,a previously proposed mechanism for MCR,including an organometallic methyl-Ni(III)F430intermediate,has beenruled out,and a new mechanism involving a Ni(II)-SCoM intermediate and a methyl radical was proposed.Other possible competitive mechanisms were also investigated.
