Highly efficient synthesis of imidazo-fused heterocycles via two multicomponent reactions catalyzed

  Multicomponent reactions (MCRs) have emerged as powerful tools to construct diverse functionalized molecules via the formation of multiple bonds in a one-pot operation with high atom efficiency,straightforward reaction which accord with the principles of green chemistry.MCRs can quickly construct chemical molecule libraries of drug-like heterocycles with target-oriented functionalities.Among these various heterocyclic derivatives,imidazo-fused heterocycles exhibit a broad spectrum of biological activities.Two simple,efficient and eco-friendly one-pot three-component reactions for the convenient synthesis of imidazo-fused heterocycles has been developed,which was catalyzed by PrCl3 in ethanol at 60℃ via condensation of aldehydes,aminoazines,isocyanides or trimethylsilyl cyanide.A wide range of imidazo-fused heterocycles were synthesized with high yields,shorter reaction time and more simple purification procedure and byproduct was successfully separated.In addition,starting from the product of Groebke-Blackburn-Bienayme (GBB) reaction,a class of novel polycyclic heterocycles was obtained with a Pd(OAC)2-catalyzed coupling reaction.
