Circulating tumor cells,which disseminate from primary tumors and circulate in bloodstream,represent a liquid biopsy to non-invasively monitor cancer metastasis1.In this work,we introduced an integrated microfluidic system for fast and efficient circulating tumor cell (CTC) isolation and capture.The microfluidic platform consists of a combination of microfluidic deterministic lateral displacement (DLD) array2 and affinity-based cell capture architecture3.The microfluidic DLD array enables fast and efficient circulating tumor cell isolation based on cell size differences,and the affinity-based cell capture structure allows for continuous high throughput and purity cancer cell capture.Using this device to isolate MCF-7 breast cancer cell from spiked blood samples,we achieved an enrichment factor of 1500X,and a high processing throughput of 9.6 mL/minh with 90% capture yield and more than 50% capture purity.We also demonstrate the capability of the integrated system for cancer cell culture and drug screening.Cancer cells were first captured onto antibody modified substrate surface and cultivation,then the analysis of different anti-cancer drug effect on cells was performed.This integrated platform offers a promising approach for CTC capture with high recovery rates,purity and stability.The system demonstrates unique capability for fast and efficient CTC detection and characterization,and the on-chip drug screening exhibits great potential for assessment of personalized medicine.
