The pharmaceutical development is greatly hindered by the poor predictive power of existing animal testing and cell-based models.Development of a human-based in vitro system has the potential to make better predictions of human response to drugs.Organs-on-chip technology can recapitulate many key aspects of the human tissue and organ physiology in a physiologically relevant manner, which is well suited for the development of assays in drug efficacy and toxicity testing in vitro.In this talk, I will present our efforts to design the multi-scale organs-on-a-chip that can be used for the purpose of disease modeling and drug toxicity testing in normal and pathological states, such as in liver, heart and brain diseases.The properties and functions of these microdevices for mimicking the microphysiological system are described.The design processes with the attention of particular cells, biochemical and biophysical cues and are also presented.We hope this technology has great potentials to advance our understanding in translational toxicology.
