Traditional strategies of preparing photo-responsive block copolymer(BCP)micelles as drug carriers often involve toxic photo-cleavable moieties.To explore a new field in chemistry for more controllable,tunable,effective and safer drug delivery,we developed a novel simultaneously photo-cleavable and activatable(SPCA)prodrug backboned BCP micelle strategy using anticancer prodrugs,Pt(Ⅳ)-azide complexes as both polymer backbone building bricks and light-responsive groups.Upon the application of UV light or even visible light,the Pt(Ⅳ)prodrug is cleaved to its active form,Pt(Ⅱ)drug,and the active drug gets released.The use of the inert prodrug could conceal the cytotoxicity of the anticancer drug against normal cells and circumvent the toxic side effects caused by photochromic groups and small molecular byproducts.The excellent triggerinduced anticancer ability and toxic side effect remission ability of SPCA prodrug backboned BCP micelles were also proven in vitro and in vivo.
