Time-lapse imaging of quail embryonic hearts was performed to observe the morphogenesis of the myocardial and endocardial heart tubes.Use of Tiel+ transgenic quail (courtesy of Dr.R.Lansford, Caltech) and electroporated nuclear markers were used to follow myocardial and endocardial precursor cells from gastrulation stages through early stage heart looping.In contrast to published data on cardiac development, time-lapse movies revealed a continuous, circular primary heart field exists in avians, and indicated the stages at which anterior-posterior patterning of the myocardial precursor cells was established.Further, comparison to in situ hybridization data using well-known cardiac markers revealed some myocardial precursor cells do not express Nkx2.5 until Stage 10, when the tubular heart is already formed.
