  Genotyping of gene-linkage or gene-target markers was the most crucial part of predicting (or diagnosing) whether desired genes exist in germplasm and progenies in crop molecular breeding procedure such as MAS (marker-assisted selection).It has become a major bottleneck that preparation of sufficient quantity and good quality genomic DNA from various tissues of large number of samples of breeding population in rice.A variety of DNA preparation methods and commercial kits are available.However,they are either time-consuming,low throughput,low yield,or costly.The other,emerging analytical system such as HRM,T-ARMS-PCR and KASPTM which were based on SNP (single nucleotide polymorphism) had been applied increasingly,besides SSR and InDel analyses.These necessitate the development of a universal,more efficient and labor-saving protocol for isolating compatible gDNA from rice plants,and being applied to molecular breeding.
