Glucosaminoglycans (GAG) and particularly Heparan sulfates (HS) serve multiple roles in tissue architecture and homeostasis and multiple proteins share unique HS binding domains.Twenty years ago I have pointed out that HS may have a critical role in regulating the specific activity of fibroblast growth factors (FGF), by directly modulating their binding and activation of their cognate cell surface receptors (Yayon et al.Cell, 1991).With time, the role of HS as physiological regulators of multiple growth factors and cytokines during embryonic development, tissue repair and regeneration has been demonstrated and the unique function of protein GAG interactions of both ligands and their receptors is currently widely accepted.
