OBJECTIVE: To improve the temperature sensitive release properties of the novel thermosensitive liposomes(NTSL) composed of DPPC and HSPC.METHODS: Calcein-loaded TSLs with different phospholipid compositions were prepared by the thin film-hydration method.Based on the fluorescence self-quenching off characteristics of calcein, the release behavior of the above liposomes was compared when exposed to 37 ℃ (body temperature) as well as phase transition temperature (Tm) of TSL.Thus prescription of NTSLs was optimized and the temperature-sensitive release properties of resultant liposomes in different release media were investigated in comparison with DPPC TSLs.RESULTS: Compared with the traditional DPPC TSLs liposomes, the NTSL showed more favorable temperature-sensitive release performance and more stable encapsulation ability to calcein, even if the amount of lysophospholipids was reduced by 60%; The release of in mouse plasma is similar to that in human plasma ; The results of tissue distribution study revealed that ,the NTSL had improved targeting drug delivery properties.CONCLUSIONS : NTSLs with higher Tm possess significantly improved temperature-sensitive release properties.
