Cells navigate environments and regulate gene expression in response to specific signals.Engineers seek to harness this capability to reprogram cells to perform complex tasks or create useful materials.Nuclear factorkB (NF-κB) is a kind of inducible transcription factor, which regulates gene expression by binding to specific DNA sequences, known as κB sites.The unique DNA-binding properties of different dimers in NF-κB family regulate the specific expression of target genes.To more thoroughly investigate these dimer-specificdifferences, we use FACS(Fluorescence Activated Cell Sorter) to evaluate DNA sites recognized by different NF-κB dimers in yeast.We observed some specific regulatory κB sites and build some basic regulatory elements.In the future we will try to uncover the regulatory dynamics of κB sites and build more complex circuits.This study provides a database that can be used to identify NF-κB target sites in yeast and in human T cell and be used as basic synthetic biology parts and helps illustrate the complex mechanisms of transcriptional regulation in eukaryotes.
