Preparation and physico-chemical property of Panax notoginseng saponins liposomes

  Purpose: In this study, Panax notoginseng saponins (PNS) was encapsulated in small unilamellar vesicles (SUVs) and the effects of formulation variables on the liposome characteristics were investigated.Methods: Liposomes were prepared using ethanol injection method.A number of variables such as molar ratios of phospholipid (hydrogenation phospho lipid, HSPC ; soybean phospholipid, PC), cholesterol, PNS, α-tocopherol and phosphate buffered saline were studied.In vitro characterization of the liposomes including the encapsulation capacity,size and Zeta potential were carried out.Results: The ultimate of preparation formula was as follows:the absolute ethanol was used as the solvent; α-tocopherol was used as the antioxidant; the ratio of HSPC to PC (w/w) was 1.5 ∶ 1, while the ratio of cholesterol to lipid (molar ratio) was 1 ∶ 3, the drug-lipid ratio (w/w) was 1 ∶ 30.The characters of the prepared liposome was investigated ,which gave us the mean diameter of 133 nm, Zeta potential of-23.0 mV.The encapsulation efficiency of the liposome determined by Sephadex G-50 was 39.7%.Stability test show that the liposome suspension was stable at the temperature 4±1℃ for 3 months.Conclusion: The ethanol injection method is a simple and efficient technique for designing liposomes for water-soluble PNS.
