Lansoprazole (LAN), a selective proton pump inhibitor, has a chiral benzimidazole sulfoxide structure and is used for the treatment of gastric acid hypersecretory related diseases.In order to investigate its stereoselective pharmacokinetics, a stereoselective and high through-put UPC2-ESI-MS/MS method was developed for simultaneous quantification of LAN enantiomers in dog plasma.After protein precipitation with acetonitrile, LAN enantiomers and internal standard (pantoprazole enantiomers) were separated on a Waters Acquity Trefoil CEL1 column using Waters UPC2 system.The mobile phase was 0.1% formic acid methanol: CO2 (20∶ 80, V/V) at a flow rate of 2.0 ml/min.The column oven temperature was 55℃ and the ABPR pressure was 2500 psi.
