目的:研究复方丹参滴丸对人肝脏内黄嘌呤氧化酶(XO)活性的影响,从而指导临床合理用药。方法:采用反相高效液相梯度洗脱法测定服用复方丹参滴丸前后人尿液内咖啡因4种主要代谢产物5-乙酰氨基-6-甲酰氨基-3-甲基尿酸(AF-MU)、1-甲基尿酸(1U)、1-甲基黄嘌呤(1X)和1,7-二甲基尿酸(17U)的相对含量,采用1U/(1X+1U)评价人肝脏内XO活性的变化。结果:受试者用药前XO平均活性为(0.33±0.05),服用复方丹参滴丸14d后XO平均活性为(0.32±0.07),服用复方丹参滴丸28d后XO平均活性为(0.31±0.04),XO的平均活性在服药中期、服药后分别比服药前降低3.03%,6.06%,服药中期与服药前相比较差异无显著性(P>0.05),服药后期与服药前相比较,XO活性降低,差异有显著性(P<0.05)。结论:长时间服用复方丹参滴丸对XO活性有一定的抑制作用,在与经XO代谢的药物联合应用时会影响其药物疗效。
Objective: To study the effect of Compound Danshen Dripping Pills on the activity of xanthine oxidase (XO) in human liver, so as to guide clinical rational drug use. METHODS: A reversed-phase high performance liquid phase gradient elution method was used to determine the four main metabolites of caffeine in human urine, 5-acetamido-6-carboxamido-3-methyluric acid (AF-MU) before and after taking compound Danshen dripping pills. The relative amounts of 1-methyluric acid (1U), 1-methylxanthine (1X), and 1,7-dimethyluric acid (17U) were used to evaluate human liver XO activity using 1 U/(1X+1U). The change. Results: The average XO activity of the subjects before treatment was (0.33±0.05). The average XO activity was (0.32±0.07) after taking compound Danshen Dripping Pills for 14 days, and the average XO activity after taking Compound Danshen Dripping Pills for 28 days (0.31±0.04). The average activity of XO was decreased by 3.03% and 6.06% before and after medication, respectively. There was no significant difference between the mid-dosage and pre-dosage (P>0.05). The XO activity was lower in the later period of medication and before the medication. The difference was significant (P<0.05). Conclusion: The long-term use of Compound Danshen Dripping Pills has a certain inhibitory effect on XO activity, and when combined with XO-metabolizing drugs, it will affect its efficacy.
