Dengue virus (DENV) causes a wide range of symptoms, from mild ferule illness, dengue fever (DF), to severe life threatening illness, dengue hemorrhagic fever (DHF) and dengue shock syndrome (DSS).A phenomenon that sub-neutralizing concentrations of antibody-dependent enhancement (ADE),which is considered to be a risk factor for DHF and DSS.In this study, rapid and sensitive assays for neutralization and ADE activity were developed using a stable Renilla luciferase reporter DENV-2.Briefly, reporter DENV-2 and serially diluted antibodies were incubated for 1 prior to culture in BHK-21 and THP-1 cells, then luciferase activity in cells was measured at 48 and 72h post-infection respectively.The RLU values showed well correlation with conventional plaque assay.This assay was further evaluated using negative and positive sera from monkey and human cases.The new assay system using report virus is sensitive, simple, prompt and reliable to measure both enhancing and neutralizing activities of antibodies.
