Neuroactive peptides that target ion channels or receptors are attractive sources of therapeutics for neurological diseases,but because of their peptidic nature,their discovery and development remain outside the mainstream interest of pharmaceutical industry.Recent advances in molecular biology and peptide engineering start to accelerate biodiversity-derived discovery of novel neurotoxins with targeted pharmacological properties.Furthermore,new repertoires of chemical modifications that improve bioavailability of neuroactive peptides are emerging.As a growing number of peptide-based lead compounds advance into preclinical and clinical studies,these peptide drugs will likely capture more attention.Two examples will be presented:(1) discovery and engineering analgesic conotoxins that block neuronal subtypes of sodium channels,and (2) developing blood-brain-barrier permeable galanin-based compounds for epilepsy and pain.These two projects will illustrate the latest developments in transforming neuroactive peptides into future neurotherapeutics.
