This article describes a fatty acid assay that uses high performance liquid chromatography (HPLC) to simultaneously detect highly sensitive saturated or unsaturated fatty acids in milk, including lauric (lauric), myristic (fourteen (Palmitic acid), stearic acid (octadecanoic acid), palmitoleic acid (palmitoleic acid), oleic acid (octadecenoic acid), and linoleic acid Carbonic acid). Fatty acids were first derivatized with 2- (2-naphthyloxy) ethyl 2- (piperidinyl) ethanesulfonate (NOEPES) and their naphthyloxyethyl derivatives were subjected to isocratic elution by reverse phase HPLC. The fatty acids in milk are extracted with toluene and used directly in NOEPES derivatization without the need for solvent replacement. Fatty acid content was measured by standard addition method. This method requires only a small amount of milk sample (10 μL) to complete the analysis test.
