To construct pcDNA3.1-VEGF165plasmid,and observe its effect on repair of rabbit bone defects.Total RNA was extracted from rabbit tissue.cDNA was prepared by inverse transcription and the gene was cloned by PCR.Plasmid pMD18-T/VEGF165 combined with pcDNA3.1 was cloned to reconstruct pcDNA3.1/VEGF165 plasmid.Twenty-eight New Zealand white rabbits weighing 2.5±0.13 kg were used to make a model of bone defect (1 cm in length) of bilateral radii.The skin of the bilateral radii was cut down and 1 cm bone in the middle of radius was resected.The defects were repaired with absorbable gelatin sponge.Local injection of sodium chloride was done in the control group,and local injection of rabbit rercombinant VEGF165 0.2 ml (200ng) in the experimental group.
