Objective: To investigate the change of proteomics expression in human bone marrow mesenchymal stem cells introduced by hydroquinone and to screen for the target protein.Methods: Isolate and cultivate the human bone marrow mesenchymal stem cells till the third generation, then divide them randomly into two groups: the experiment group and the control group.In the experiment group, the bone marrow mesenchymal stem cells were exposed to hydroquinone at 5x10-5mol/L for 24 hours while in the control group, the cells were treated with the same concentration phosphate buffered solution.The total protein was isolated and purified after exposure.The two dimensional gel electrophoresis and mass spectrum were used for detecting the changes in proteomics expression, and then find the target protein hydroquinone affect.Results: Among 3 repeated experiments, each have about 20 different proteins between the control group and the hydroquinone treated group.But there are only 11 well repeated protein sports, in which Annexin-Ⅱ, Formyl-glutathione hydrolase, UDP-glucose dehydrogenase and Aldolase A protein were identified by LC-Qq-TOF MS/MS.Conclusion: Hydroquinone exposure can decrease Annexin-Ⅱ, Aldolase A, UDP-glucose dehydrogenase, while increase Formyl-glutathione hydrolase protein.
