Grapes are one of the most widely cultivated fruit crops in the world.Chinese wild grape germplasm are important resources for crossbreeding and molecular breeding.Vitis quinquangularis is a unique germplasm with high resveratrol content,disease-resistance and has been well documented.However,the changes of its proteins and metabolites profiles during berry development and ripening are not clear yet.The aim of the present study was to investigate changes in the berry skin proteome/transcriptiome during ripening in Vitis quinquangularis accession Danfeng-2 and comparative proteomes/transcriptiome were conducted with a low resveratrol content variety,namely,Vitis vinifera cv.Cabernet Sauvignon.For proteome analysis,8 × plex of isobaric tags for relative and absolute quantification(iTRAQ)MS/MS was used to generate markedly changes in the berry skin proteomes.A total of 3751 proteins were detected and differentially expressed proteins across four key time points or interspecies were classified.Proteins obtained were analyzed to explore their potential functions.Several proteins involved in Phenylalanine metabolism pathway were differently expressed between Danfeng-2 and Cabernet Sauvignon at version.The competition between chalcone synthase branch and stilbene synthase branch were verified.In addition,there were various proteins with conserved expression profiles during berry ripening in Danfeng-2 and Cabernet Sauvignon.RNA-seq analysis was conducted on an Illumina HiSeq 2500 instrument.Genes involved in phenylalanine metabolic pathway were selected and combined with corresponding proteins identified in iTRAQ experiment.Single Nucleotide Polymorphisms(SNP)and alternative splicing events were also analyzed.Furthermore,novel genes from Danfeng-2 were assembled and annotated.Finally,a global view of the compared omics between varieties were displayed.
