Gene expression is a risk-taking process at the presence of cellular post-transcriptional gene-silencing(PTGS)machineries.However,it is unclear why and how most endogenous coding transcripts avert a fate of PTGS.To understand the regulation of PTGS,we firstly performed a cosuppression-based suppressor screen in Arabidopsis and obtained three genes essential for either 5 or 3 cytoplasmic mRNA decay.Interestingly,we found that the 5 and 3 cytoplasmic mRNA decay pathways had intensive genetic interactions and showed a synthetic-lethality phenotype that was primarily due to genome-wide PTGS events.
