目的:观察降糖三黄片对2型糖尿病大鼠胰岛β细胞BIP基因表达的影响,探讨其干预内质网应激介导的β细胞凋亡的作用机制。方法:40只4周龄Wistar大鼠被随机分为正常组和2型糖尿病组。高脂+蛋氨酸饲料喂养30只低周龄Wistar大鼠1个月,建立胰岛素抵抗模型,结合小剂量STZ腹腔注射,建立2型糖尿病大鼠模型,随机分为2型糖尿病模型组和降糖三黄片组,降糖三黄片组予以灌胃中成药降糖三黄片治疗2个月,测定血清胰岛素、血脂、血糖,进行治疗前后比较;免疫组化方法测定各组大鼠胰岛β细胞BIP基因的表达。结果 :降糖三黄片可明显改善2型糖尿病大鼠血糖、血脂代谢紊乱,改善胰岛素抵抗,明显上调内质网应激特征性分子BIP基因的表达。结论:降糖三黄片通过上调2型糖尿病大鼠胰岛β细胞BIP基因的表达,维持胰岛β细胞内环境的稳定,保护胰岛β细胞,减少内质网应激介导的胰岛β细胞凋亡。
Objective: To observe the effect of Jiangtang Sanhuang Tablet on the expression of BIP gene in pancreatic β cells of type 2 diabetic rats, and to explore the mechanism of its intervention on endoplasmic reticulum-induced β-cell apoptosis. Methods: Forty Wistar rats of 4 weeks old were randomly divided into normal group and type 2 diabetic group. 30 low-Wistar rats were fed with high-fat + methionine for 1 month to establish model of insulin resistance, combined with low-dose STZ intraperitoneal injection to establish type 2 diabetic rat model and randomly divided into type 2 diabetes model group and hypoglycemic three Huangpi group, hypoglycemic Sanhuang tablet group was given intragastric administration of Chinese medicine hypoglycemic Sanhuang tablets for 2 months, serum insulin, blood lipid, blood glucose were measured before and after treatment; immunohistochemistry was used to measure the islet β-cell BIP gene expression. Results: Jiangtang Sanhuang Tablet can significantly improve blood glucose and lipid metabolism disorder, improve insulin resistance and up-regulate the expression of BIP gene in endoplasmic reticulum stress-induced type 2 diabetic rats. Conclusion: Jiangtang Sanhuang tablet can up-regulate the expression of BIP gene in pancreatic β-cells in type 2 diabetic rats and maintain the stability of pancreatic β-cells, protect the islet β cells and decrease the endoplasmic reticulum stress-induced pancreatic β-cell apoptosis.
