目的 研究夏枯草三萜酸对大鼠急性肝损伤的预防保护作用及其可能机制.方法 建立CCl4诱导大鼠急性肝损伤模型,将雄性SD大鼠60只随机分为正常组、模型组、夏枯草三萜酸62.5,125和250 mg· kg-1及联苯双酯0.2 g·kg-1组,每组10只,灌胃给药6d,末次给药1h后除正常组外一次性腹腔注射CCl4(0.1 ml·100 g-1),造成急性肝损伤模型.禁食不禁水16 h,腹股动脉取血,处死大鼠,剖取肝脏.观察夏枯草三萜酸对大鼠血清中丙氨酸氨基转换酶(ALT)、天门冬氨酸氨基转换酶(AST)水平的影响以及对肝匀浆中丙二醛(MDA)、超氧化物歧化酶(SOD)水平的影响,计算肝指数并同时对肝组织进行病理学检查,采用RT-PCR,Western blot法检测组织中CYP2E1表达变化.结果 夏枯草三萜酸各剂量组可减弱急性肝损伤大鼠血清中ALT,AST的活性,降低肝匀浆MDA的水平,升高SOD的水平,此外,RT-PCR,Western blot结果显示夏枯草三萜酸能明显降低大鼠体内CYP2E1表达,呈剂量依赖关系.结论 夏枯草三萜酸对CCl4致大鼠急性肝损伤具有明显的保护作用,其机制可能与抗脂质过氧化有关.
